Appendix 1 Calberlas staining solution Materials 5ml glycerine Method Mix together a 50:50 solution of fuschin and glycerol. Upon cooling the solution will form a dark pink gel. Store in an airtight jar for future use. Mix the glycerine, ethanol, distilled water and 6 drops of the melted fuschin/glycerol solution. Store the Calberlas solution in a clear bottle with a dropper lid. Place a drop of Calberlas on the slide and cover with a coverslip. Allow ~ 10 minutes before observing pollen structure under the microscope. Calberlas stains pollen grains pink, but not fungus spores nor most of the other debris usually present on samples exposed outdoors. The dye is deposited on the outer wall of pollen grains, enhancing the surface characteristics necessary for identification. Basic fuschin is available from a number of suppliers. It comes from the supplier in a powder form. Only a very small amount is needed to make up the saturated aqueous basic fuschin. Suppliers:
